Nuritas Published Peer-Reviewed PeptiSleep™ Sleep Study in Sleep Medicine: X

Peer-reviewed study pairs clinically validated questionnaires with Oura Ring biometrics, offering a solid blueprint for how functional ingredients will be substantiated in a wearable-first world

DUBLIN, Ireland – [August 11] – Nuritas, the AI-driven biotech company that discovers and clinically validates bioactive peptides from natural sources, today announced the publication of a peer-reviewed pilot study on its sleep ingredient PeptiSleep™ in Sleep Medicine: X, the open-access companion journal to Sleep Medicine, published by Elsevier.

The study, “Effects of PeptiSleep™ on Sleep Modification and Biometrics in Adults with Sleep Impairment: A Randomised, Placebo-Controlled Pilot Study,” is notable not only for its findings on PeptiSleep™ itself, but for how it was measured: alongside the clinically validated Leeds Sleep Evaluation Questionnaire (LSEQ), every participant wore an Oura Ring throughout the trial to passively track over 15 biometrics including heart rate variability, sleep architecture, and sleep latency in real-world, at-home conditions, not a sleep lab.

Study design and findings

The 8-week (56-day), randomised, placebo-controlled, parallel-design pilot enrolled 26 adults (aged 18–65) with self-reported sleep impairment. Participants received either PeptiSleep™, a rice protein hydrolysate containing AI-discovered bioactive peptides derived from brown rice, or a placebo.

Key findings:

  • First AI-discovered sleep ingredient tested in a placebo-controlled trial: PeptiSleep™, a rice protein hydrolysate identified using Nuritas’ AI discovery platform, is the first in silico-predicted plant peptide ingredient to be assessed in a placebo-controlled sleep trial.
  • Subjective and objective data on sleep quality were consistent:PeptiSleep™ users reported improvements across all four domains of the LSEQ (ease of getting to sleep, quality of sleep, behavior following wakefulness, and alertness on waking), with nominally significant differences versus placebo in the sleep initiation(p=0.050) and sleep maintanance (p=0.036) domains. This was supported by trends in exploratory objective data collected using the Oura wearables showing higher heart rate variability, lower nocturnal heart rate, and deep sleep, signals of more restorative sleep.
  • Mood improved alongside sleep, with a strong safety profile: The PeptiSleep group showed a nominally significant reduction in total mood disturbance (p=0.029) versus placebo by day 56, and the supplement was well tolerated over the 8-week study with no clinically significant safety findings
  • Findings from the exploratory pilot with dynamic trial design warrants follow-on trial: As a placebo-controlled pilot study (n=26), these findings are exploratory, directional, and hypothesis-generating rather than confirmatory. That said, the consistency across independent measures, subjective questionnaires, mood scales, and passive wearable biometrics, gives Nuritas a strong signal to build on. The team is now planning larger, follow-on trials to build on these results
